FYN

FYN全称为酪氨酸蛋白激酶Fyn,属于Src家族成员,在细胞信号传导中发挥核心作用。FYN存在两种异构体,分别通过不同剪切方式产生p59-Fyn和p58-Fyn,在T细胞受体激活、神经元发育等过程中参与调控。其异常活化与多种恶性肿瘤密切相关,Fyn激酶在乳腺癌、前列腺癌中通过PI3K/AKT通路促进肿瘤转移,在胶质母细胞瘤中与表皮生长因子受体形成致癌复合物。阿尔茨海默病患者脑组织中发现Fyn与Tau蛋白异常互作,加速神经纤维缠结形成,而帕金森病模型中也观察到该激酶对α-突触核蛋白聚集的促进作用。针对Fyn的药物开发聚焦于激酶抑制剂领域,达沙替尼等广谱Src抑制剂已进入临床研究,新型选择性抑制剂如AZD0424在临床前实验中显示出对三阴性乳腺癌的特异性抑制作用。近期研究还发现Fyn在免疫检查点调控中的作用,其与PD-1受体的相互作用为肿瘤免疫治疗提供了新靶点。

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
