CCR3

CCR3(C-C趋化因子受体3)是一种主要分布于嗜酸性粒细胞、嗜碱性粒细胞及Th2细胞的G蛋白偶联受体,也被称为CD193。该蛋白通过与Eotaxin等趋化因子结合,调控免疫细胞向炎症部位迁移,尤其在过敏性疾病中扮演核心角色。研究发现,其在哮喘患者的呼吸道、过敏性鼻炎鼻黏膜及特应性皮炎皮损部位异常活跃,与嗜酸性粒细胞浸润直接相关。近年还发现某些实体瘤微环境中CCR3高表达,可能参与肿瘤免疫逃逸,例如结肠癌中CCR3信号可能促进血管生成。

针对CCR3的药物研发曾聚焦于小分子拮抗剂,如GSK766994等早期候选药物在临床试验中因疗效不足或脱靶效应未能突破。目前策略转向联合疗法,例如将CCR3抑制剂与抗IgE抗体奥马珠单抗联用,或开发双功能抗体同时阻断CCR3与IL-5通路。2022年跨国药企公布的临床前数据显示,新型变构抑制剂可选择性抑制嗜酸性粒细胞迁移而不影响其他免疫细胞,为减少副作用提供新方向。但该靶点成药性挑战仍存,需进一步验证其在复杂炎症网络中的调控机制。
