CD14

CD14蛋白全称为单核细胞分化抗原CD14,也被称作MY23、LeuM3或p55,是一种锚定在细胞膜表面的糖蛋白,主要分布于单核细胞、巨噬细胞及中性粒细胞表面。CD14作为脂多糖(LPS)的高亲和力受体,能够识别并结合细菌内毒素,通过与TLR4受体形成复合物激活下游免疫应答。在感染过程中,CD14通过TLR4/MyD88依赖的信号通路触发NF-κB等转录因子活化,诱导TNF-α、IL-6等促炎因子释放,参与脓毒症、慢性炎症性疾病的发展。临床研究发现,CD14基因多态性与动脉粥样硬化、阿尔茨海默病及类风湿性关节炎等疾病风险相关,其表达水平在脓毒症患者血液中显著升高,已成为感染性休克的潜在生物标志物。针对CD14的抗体药物(如IC14)已进入临床试验阶段,用于中和内毒素引发的过度炎症反应,同时基于TLR4-CD14信号轴的小分子抑制剂也在探索治疗自身免疫性疾病的新策略。

IHC image of CSB-RA004879MA1HU diluted at 1:100 and staining in paraffin-embedded human liver tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
