CDK6

CDK6,即细胞周期蛋白依赖性激酶6,别名细胞分裂蛋白6、血小板来源的丝氨酸/苏氨酸激酶等。它是一种由CDK6基因编码的蛋白激酶,属于CMGC蛋白激酶家族成员,对细胞周期G1期的进展和G1/S转换具有重要作用。CDK6与D型细胞周期蛋白结合,形成复合体,通过磷酸化视网膜母细胞瘤蛋白(Rb),激活E2F转录程序,促进细胞进入S期进行DNA复制。CDK6在生物学上的意义主要体现在其对细胞周期的调控作用。它的活性对于维持细胞的正常增殖至关重要,同时CDK6也参与了细胞分化和肿瘤细胞的增殖。在肿瘤细胞中,CDK6的异常激活与多种癌症的发生发展有关,包括白血病、淋巴瘤和实体瘤等。此外,CDK6抑制剂如Palbociclib、Ribociclib和Abemaciclib已在临床上用于治疗某些类型的乳腺癌,并正在被研究用于治疗其他癌症类型。
