CX3CR1

CX3CR1全称为CX3C趋化因子受体1,也被称为Fractalkine受体或G蛋白偶联受体13。这种蛋白主要分布在单核细胞、小胶质细胞和自然杀伤细胞表面,通过与其配体Fractalkine的相互作用,调控免疫细胞的迁移与黏附功能,尤其在炎症反应和免疫监视中发挥重要作用。研究发现,CX3CR1介导的细胞间通讯与神经退行性疾病密切相关,例如阿尔茨海默病患者脑内小胶质细胞的异常激活便与其功能失调存在关联。此外,该受体在动脉粥样硬化斑块形成过程中参与单核细胞的浸润,与心血管疾病的发展密切相关。
近年来,针对CX3CR1的药物开发聚焦于炎症性疾病治疗领域。部分小分子抑制剂已在动物模型中展现出抑制神经炎症和延缓动脉斑块进展的效果。临床前研究还发现,阻断CX3CR1可能缓解类风湿性关节炎的关节损伤。目前已有单克隆抗体类药物进入早期临床试验阶段,尝试通过调控免疫细胞迁移来治疗多发性硬化等自身免疫疾病。与此同时,该受体在肿瘤微环境中的双重作用也引起关注,既有研究支持其促进肿瘤免疫逃逸,也有证据表明可能增强抗肿瘤免疫应答,这一矛盾特性为靶向治疗带来挑战。
