CXCR2

CXCR2全称为C-X-C趋化因子受体2型,又称IL-8受体β或CD182,属于G蛋白偶联受体超家族成员。CXCR2通过与IL-8、GROα等趋化因子结合,介导中性粒细胞向炎症部位迁移,在急慢性炎症反应、组织修复及肿瘤微环境调控中发挥关键作用。CXCR2异常活化与慢性阻塞性肺病、银屑病、类风湿性关节炎等炎症性疾病密切相关,其在多种实体瘤中通过招募髓系抑制细胞促进免疫逃逸的特性也备受关注。
针对CXCR2的药物开发聚焦于小分子拮抗剂,如Navarixin(MK-7123)已完成银屑病二期临床试验,AZD5069在慢阻肺治疗中显示出抑制中性粒细胞浸润的效果。由于CXCR2拮抗可能影响正常免疫功能,部分药物因疗效不足或毒副作用终止研发。目前研究转向联合用药策略,如在胰腺癌治疗中与免疫检查点抑制剂联用,或开发双重CXCR1/CXCR2抑制剂以增强抗炎效果。该靶点的组织特异性调控机制仍是转化医学研究的难点。
