CYBB

CYBB全称为细胞色素b-245重链(Cytochrome b-245 heavy chain),又称NOX2或gp91phox,是NADPH氧化酶复合物的核心催化亚基。CYBB主要分布于中性粒细胞、单核细胞等吞噬细胞的细胞膜上,负责催化氧分子生成超氧阴离子,构成机体抵抗病原微生物的关键防线。当免疫细胞识别入侵病原体时,CYBB蛋白会与多个调控亚基组装形成活性酶复合体,通过"呼吸爆发"产生大量活性氧直接杀伤细菌,这一过程被称为氧化杀菌作用。CYBB基因突变会导致慢性肉芽肿病(CGD),患者因无法有效清除病原体而反复发生严重感染,并伴随肉芽肿形成和炎症过度反应。针对该疾病的治疗除了抗生素预防和干扰素-γ辅助治疗外,干细胞基因治疗已进入临床试验阶段,通过回输携带正常CYBB基因的造血干细胞实现功能修复。基于NOX2调控机制的抗氧化剂和抗炎药物研究也为相关免疫缺陷及炎症性疾病提供了新思路。

Immunofluorescence staining of hela cell with CSB-RA006325MA1HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Mouse IgG(H+L).
