DUSP2

DUSP2,即双特异性磷酸酶2,别名PAC1、PAC-1,是一种由人类基因DUSP2编码的蛋白质,属于双特异性磷酸酶亚家族成员。DUSP2蛋白主要在造血组织中表达,并定位于细胞核。
DUSP2的作用机制是通过去磷酸化靶激酶上的磷酸丝氨酸/苏氨酸和磷酸酪氨酸残基,从而负向调节与细胞增殖和分化相关的丝裂原活化蛋白(MAP)激酶超家族成员,如MAPK/ERK、SAPK/JNK、p38。DUSP2对ERK1和ERK2的活性具有抑制作用。
DUSP2参与调控免疫反应、炎症反应以及癌症进展,与多种疾病相关,包括血液癌症和2型糖尿病。DUSP2的功能丧失在某些癌症中预示着不良预后,并且在调控TH17细胞分化中起重要作用,影响自身免疫疾病的发生发展。
