FOSL1

FOSL1(FOS-like antigen 1)又称Fra-1,属于AP-1转录因子家族成员,在细胞增殖、分化和凋亡中发挥调控作用。FOSL1通过与JUN家族蛋白形成二聚体,结合特定DNA序列激活下游靶基因,尤其与肿瘤侵袭转移密切相关。有研究发现,FOSL1在乳腺癌、结直肠癌和肺癌等多种恶性肿瘤中异常高表达,通过诱导上皮间质转化(EMT)促进癌细胞迁移,并与血管生成、化疗耐药等恶性表型相关。其活性受MAPK/ERK和JNK等信号通路调控,与炎症因子TNF-α、IL-6的表达也存在交叉调控。目前针对FOSL1尚无直接靶向药物,但针对其上游信号分子开发的MEK抑制剂(如曲美替尼)已在临床试验中验证对部分肿瘤的疗效。近年研究发现,天然化合物姜黄素可通过抑制FOSL1表达延缓肿瘤进展,为联合用药提供了新方向。
