GNRHR

GNRHR,即促性腺激素释放激素受体,别名GRHR、Gonadotropin-releasing hormone receptor、GnRH receptor、GnRH-R。它是一种G蛋白偶联受体,主要在垂体前叶中表达,参与调节生殖激素的释放。GNRHR的作用机制主要是通过与促性腺激素释放激素(GnRH)结合,激活受体,进而通过G蛋白介导的信号转导,增加细胞内cAMP的水平,促进黄体生成素(LH)和卵泡刺激素(FSH)的释放。GNRHR不仅在生殖系统中发挥关键作用,还与多种疾病的发生发展有关。例如,在乳腺癌中,GNRHR的表达与肿瘤的临床病理特征和预后相关,特别是在激素受体阴性的乳腺癌中,GNRHR的高表达可能与改善的无病生存期(DFS)相关。此外,GNRHR的信号传导在垂体细胞中涉及丝裂原活化蛋白激酶(MAPK)级联的激活,为信号从细胞表面到细胞核的传递提供了重要的联系,并在促性腺激素的转录调节中起作用。GNRHR也在非垂体细胞中表达,并发现其在某些细胞中抑制细胞增殖,因此,GNRHR代表了用于癌症治疗的GnRH类似物的潜在靶标。
