HCN1

HCN1,即超极化激活型环核苷酸门控通道1,别名If、Ih、HCN1等。它是一种由HCN1基因编码的膜蛋白,属于超极化激活型环核苷酸门控通道家族成员,在心脏和大脑等组织中表达。HCN1的作用机制是在细胞膜超极化时激活,允许钠和钾离子通过,对心脏节律和神经元兴奋性具有调节作用。HCN1参与了心脏的自主节律控制和心脏起搏,同时在神经系统中调节神经元的发放频率和信号传递。HCN1的功能异常与心脏疾病中的窦性心动过缓、长QT综合征,以及神经系统疾病中的癫痫有关,是心脏学和神经生物学研究中的一个重要分子靶点。

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunofluorescence staining of HepG2 cells with CSB-PA010216LA01HU at 1:166, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
