HSPA5

HSPA5,即热休克蛋白家族A成员5”,别名BiP、GRP78。它是一种分子伴侣蛋白,主要在内质网中发挥作用,由人体细胞自身的基因编码。HSPA5的作用机制涉及帮助蛋白质的正确折叠、组装和运输。当细胞受到各种应激条件,如热休克、氧化应激或营养物质缺乏时,HSPA5的表达会显著增加,以应对蛋白质折叠错误和聚集的问题。HSPA5通过形成复合体与未折叠或错误折叠的蛋白质结合,防止它们聚集,并促进其正确折叠或重折叠。HSPA5在生物学上的意义主要体现在其对细胞应激反应的调控和蛋白质稳态的维持上。作为内质网应激反应的关键分子伴侣,HSPA5参与了未折叠蛋白反应(UPR),在细胞适应和生存中发挥重要作用。HSPA5还与多种细胞过程相关,包括细胞信号传导、细胞周期控制、细胞凋亡以及免疫应答。在肿瘤细胞中,HSPA5的高表达与肿瘤细胞的增殖、侵袭和化疗抗性有关,使其成为癌症治疗中的潜在靶点。此外,HSPA5在神经退行性疾病中也有重要作用,其功能异常可能与疾病的发生发展相关。

IHC image of CSB-RA847116A0HU diluted at 1:100 and staining in paraffin-embedded human breast cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
