HTRA1

HTRA1全称为高温需求丝氨酸蛋白酶A1，也被称为PRSS11或ARMD7。这种蛋白由丝氨酸蛋白酶结构域和PDZ结构域组成，能够通过切割细胞外基质成分及调控转化生长因子β（TGF-β）等信号分子，影响血管生成和组织重塑过程。HTRA1的异常表达与年龄相关性黄斑变性（AMD）密切相关，其基因突变可导致湿性AMD患者脉络膜新生血管过度增生。HTRA1在脑小血管病CARASIL综合征中也扮演关键角色，患者常因HTRA1功能缺失引发脑白质病变及早期卒中。
近年来针对HTRA1的药物研发聚焦于眼科疾病领域。例如Ionis公司开发的HTRA1靶向反义寡核苷酸IONIS-HTTRx已进入II期临床，用于治疗地理萎缩型AMD。部分单抗类药物通过抑制HTRA1与血管内皮生长因子（VEGF）的协同作用，试图突破现有抗VEGF疗法的耐药性问题。在肿瘤领域，由于HTRA1在胰腺癌、卵巢癌中呈现高表达特征，其蛋白酶活性调控机制也成为潜在治疗靶点。

IHC image of CSB-RA835695MA1HU diluted at 1:200 and staining in paraffin-embedded human placenta tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
