ITGA5

整合素α5(ITGA5)是细胞表面粘附分子家族的重要成员,常与β1亚基结合形成α5β1异源二聚体受体。ITGA5因能特异性识别纤维连接蛋白中的RGD序列,被称作纤维连接蛋白受体,在胚胎发育、创伤修复等生理过程中调控细胞粘附与迁移。α5β1通过激活细胞内FAK/Src信号网络,促进肿瘤细胞侵袭转移能力,其异常表达与乳腺癌、肝癌、结直肠癌等恶性肿瘤的进展密切相关。在特发性肺纤维化、动脉粥样硬化等疾病中,ITGA5也参与异常细胞外基质沉积过程。目前针对α5β1的靶向药物已有十余种进入临床研究,如单抗药物Volociximab在卵巢癌治疗中显示出抑制血管新生的潜力,小分子抑制剂JSM6427则应用于眼底黄斑病变的干预。由于整合素家族存在功能代偿效应,部分药物在临床试验中出现疗效波动,如何精准调控其活性仍是研发难点。

IHC image of CSB-RA011868MA1HU diluted at 1:50 and staining in paraffin-embedded human placenta tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
