黑色素瘤抑制性活性蛋白(Melanoma inhibitory activity,MIA)又称MIA1或CD-RAP,最初在黑色素瘤细胞中发现,属于分泌型胞外基质蛋白家族。MIA通过结合细胞表面整合素受体(如α4β1、α5β1)干扰细胞与基质的黏附,同时激活ERK/MAPK信号通路,促进肿瘤细胞侵袭转移。MIA在黑色素瘤、乳腺癌、肝癌等多种恶性肿瘤中高表达,其水平与肿瘤分期、转移风险正相关,可作为预后标志物。近年来,针对MIA的靶向治疗策略逐渐兴起,德国团队开发的MIA抗体已进入临床前研究,能有效抑制黑色素瘤细胞迁移。小分子抑制剂如PD98059通过阻断MIA下游信号通路显示出抗肿瘤潜力。非癌性疾病中,MIA还参与软骨发育调控,其基因突变与骨骼发育异常相关。
