MRAS

MRAS全称为肌肉RAS癌基因同源物(Muscle RAS oncogene homolog),属于RAS超家族成员,也被称为RAS样蛋白12(RASL12)。这类蛋白作为细胞内信号转导的"分子开关",通过结合GTP或GDP状态调控下游通路,尤其在胚胎发育、细胞周期调控中扮演关键角色。MRAS异常激活与多种实体瘤密切相关,例如非小细胞肺癌中检测到该基因的特异性突变位点,可能通过干扰细胞分化程序促进肿瘤进展。MRAS在心脏肥大反应和神经发育障碍中也显示出调控作用,其表达异常与努南综合征等遗传性疾病存在关联。
针对MRAS靶点的药物开发长期受限于其蛋白结构的特殊性,传统小分子抑制剂难以有效结合。但随着KRAS G12C变构抑制剂的成功,研究者开始探索针对MRAS特有突变(如G23V)的共价结合策略。目前已有数款候选化合物进入临床前研究阶段,通过模拟天然蛋白构象或阻断效应蛋白结合域来干预信号传导。MRAS与SHOC2-PP1C复合物的特异性相互作用为开发精准疗法提供了新方向,相关研究成果已进入转化医学验证阶段。
