POLE
POLE,即DNA聚合酶ε,别名POLE1。POLE基因编码的DNA聚合酶ε是DNA复制过程中的关键酶,它负责在DNA复制叉处添加新的核苷酸,并具有校对功能,以确保复制的准确性。POLE的作用机制涉及其聚合酶活性和校正活性,其中校对功能通过其外切酶域实现,能够识别并切除错误的核苷酸配对。POLE基因突变,尤其是外切酶域的突变,会导致DNA复制过程中的错误不能被及时纠正,从而引起高突变负荷(hypermutagenesis),增加肿瘤发生的风险。POLE突变与多种肿瘤的发生发展有关,包括子宫内膜癌、结直肠癌、非小细胞肺癌等。POLE突变型肿瘤因其高突变负荷和可能产生的新抗原而成为免疫治疗的潜在候选者。POLE突变检测在肿瘤的诊断、预后评估和治疗选择中具有重要意义。
