PSENEN

PSENEN,全称为早老素增强子γ-分泌酶亚基(Presenilin enhancer gamma-secretase subunit),别名PEN-2、MDS033。PSENEN是γ-分泌酶复合体的核心组成之一,与早老素(PSEN1/PSEN2)、APH-1等蛋白共同参与跨膜蛋白的切割过程,例如淀粉样前体蛋白(APP)经其剪切后产生β淀粉样蛋白,这类蛋白异常沉积与阿尔茨海默病的病理特征密切相关。PSENEN通过调控Notch受体的活化影响细胞分化与发育,其功能异常可能引发家族性黑蒙痴呆等罕见遗传病。近年来研究发现,PSENEN基因突变还与某些皮肤肿瘤的发生相关。在药物研发领域,针对γ-分泌酶复合体的抑制剂(如Semagacestat)曾进入阿尔茨海默病临床试验,但因干扰Notch通路引发严重副作用而终止。目前研究转向开发选择性调节剂,试图在减少β淀粉样蛋白生成的同时保留Notch信号功能,相关化合物如Nirogacestat已在乳腺癌和硬纤维瘤领域开展II期试验。

IHC image of CSB-PA878932OA01HU diluted at 1:400 and staining in paraffin-embedded human adrenal gland tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
