STAT3

STAT3,即信号转导和转录激活因子3,别名包括急性调节蛋白、信号转导激活因子、亚油酸诱导蛋白等。STAT3是一种转录因子,广泛存在于多种细胞类型中,由人体细胞自身的基因编码。其作用机制主要是在细胞接收到外部信号,如细胞因子、生长因子或其他刺激后,被激活的酪氨酸激酶磷酸化,形成二聚体,随后转移到细胞核内调控特定基因的表达。STAT3的激活涉及JAK/STAT、IL-6/STAT3等信号通路,其中Janus激酶(JAK)首先被激活并磷酸化STAT3,导致其同源二聚化和核内转位。STAT3在生物学上的意义非常广泛,它参与调节细胞的多种生理过程,包括细胞增殖、分化、迁移、凋亡以及免疫应答等。STAT3的激活对于维持细胞稳态和适应外界变化至关重要。此外,STAT3在多种疾病的发生发展中也扮演着关键角色,尤其是在肿瘤发展中,STAT3的持续激活与肿瘤细胞的增殖、存活、血管生成和免疫逃逸等密切相关。因此,STAT3成为了癌症治疗中的一个重要靶点。在自身免疫性疾病、炎症反应和组织修复中,STAT3的调节作用同样不容忽视,靶向STAT3的干预策略可能为这些疾病的治疗提供新的途径。

The cells were fixed with 70% Ethylalcohol (18h) and then incubated in 10% normal goat serum to block non-specific protein-protein interactions followedby the antibody (1µg/1*106 cells) for 1 h at 4℃.The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 30min at 4℃. Control antibody (green line) was Rabbit IgG (1µg/1*106 cells) used under the same conditions. Acquisition of >10,000 events was performed.
