TIGIT

TIGIT,即T细胞免疫球蛋白和ITIM结构域蛋白,别名WUCAM、Vstm3、VSIG9。TIGIT由人类TIGIT基因编码,该基因位于3号染色体的q13.33区域。TIGIT是一种抑制性受体,属于Ig超家族,具有细胞外的免疫球蛋白结构域、I型跨膜结构、胞内的免疫受体酪氨酸抑制/激活基序(ITIM/ITAM)。TIGIT的作用机制涉及与CD155等配体的结合,通过胞内ITIM基序传递抑制信号,减少T细胞和NK细胞的激活和增殖,降低细胞因子的产生。TIGIT在调节T细胞和NK细胞的免疫反应中起着重要作用,特别是在肿瘤微环境中,TIGIT的高表达与免疫抑制和肿瘤逃逸相关。TIGIT阻断剂在不同肿瘤模型中显示出治疗潜力,是癌症治疗的新靶点。
