VAV1
VAV1,即Vav鸟嘌呤核苷酸交换因子1,别名VAV,是一种信号转导蛋白,属于Vav家族成员之一。VAV1主要在造血细胞中表达,包括T细胞、B细胞、单核细胞、自然杀伤(NK)细胞、粒细胞和树突状细胞。
VAV1的作用机制涉及作为Rho亚家族GTP酶的磷酸化依赖性的GDP/GTP交换因子(GEF),激活导致肌动蛋白细胞骨架重新排列和转录改变的信号通路。
VAV1在T细胞和B细胞的发育和激活中发挥重要作用,参与免疫细胞生长、增殖、转化和迁移的调节。VAV1对T细胞受体激活的反应强度有重要影响,特别是钙离子参与的激活转录因子等事件。
VAV1的异常表达与多种疾病相关,包括自身免疫性疾病和肿瘤恶化,使其成为治疗和干预癌症的潜在靶点。
